This paper analyzes past and current findings of genetic causes and components of Wilms' tumor, also called nephroblastoma. The paper explains that Wilms' tumor is a kidney cancer that is thought to arise from immature cells that normally regress by birth but that occasionally persist into childhood; it is also the fourth most common type of childhood cancer. The paper points out that Wilms' tumor is historically important because it was one of the first malignancies to be treated with a combination of treatments, including surgery, radiation therapy, and chemotherapy. Citing biological studies of Wilms' tumor that laid a foundation for our understanding of how normal cells convert to cancer cells, the paper presents additional, genetic evidence that greatly adds to the understanding of how this cancer occurs. The paper concludes by noting that while While much effort has been expended on pursuing a transcriptional repressor function for WT1, it now seems that transcriptional activation is much more likely to be physiologically relevant.

"Wilms tumor, also called nephroblastoma, is a kidney cancer that is thought to arise from immature cells that normally regress by birth but that occasionally persist into childhood. Wilms tumor is historically important because it was one of the first malignancies to be treated with a combination of treatments, including surgery, radiation therapy, and chemotherapy. With this combination treatment, cure rates have risen from 40 to 50 percent in the 1960s to 85 to 90 percent today. In addition, biological studies of Wilms tumor have laid a foundation for our understanding of how normal cells convert to cancer cells. Wilms tumor is the fourth most common type of childhood cancer. Approximately 460 new cases are diagnosed each year in the United States, and 1 in 10,000 children are affected. The average age at diagnosis is about three years, although older children and even adults can occasionally develop the disease. Girls and boys are equally affected. It is uncommon for Wilms tumor to run in families; only 1.5 percent of children with Wilms tumor have an affected relative. When a family member develops Wilms tumor, it is usually a distant relative, rather than a parent or sibling -- in other words, it may be a coinci dence. For this reason we do not routinely recommend testing of the siblings of children with Wilms tumor."
